Electron probe microanalysis of the dense bodies of human blood platelets.

چکیده
Under the electron microscope various substructures were observable in the attenuated cytoplasm of platelets without any treatment, i.e., fixation, embedding or staining. The granulomere of platelets revealed dense bodies of 200-300nm diameter and smaller dense granules (about 50nm). Larger ovoid bodies with dense granules presumably corresponded to mitochondria and lysosomes. In the hyalomere there were vacuoles partly connected with surface of the platelet by a canalicule. Using an energy dispersive type X-ray microanalyzer attached to a scanning transmission electron microscope, electron probe analysis detected high levels of calcium and phosphorus and an appreciable amount of chlorine in the dense bodies of platelets. The three elements were also detected in the smaller dense granule. This technique offers some advantages for microanalytic study of the chemical components of cell organelles including the simplicity of the procedure, disuse of liquids in specimen preparation which were possibly causing substance escape, and avoidance of cryosections usually causing ice crystal artifacts.

متن کاملThe localization of calcium and phosphorus in human platelets.
Large amounts of calcium and phosphorus have been found by electron-microprobe analysis in the dense bodies of frozen-dried, human platelets. The calcium and phosphorus occurred in a fixed ratio that was similar to that in dicalcium ATP. No other elements were detectable except chlorine. Minute, electron-dense disks were found in the surface membranes of glutaraldehyde-fixed platelets, and are ...
متن کاملThe dense bodies of human platelets: inherent electron opacity of the serotonin storage particles.
SEROTONIN (5-hydroxytryptamine, 5-HT) in blood is transported almost exclusively by circulating platelets.1 Early biochemical and pharmacologic studies demonstrated that most of the 5-HT was found in granules present in platelet hyaloplasm.2” Recently, several laboratories have suggested that platelet serotonin is concentrated within a specific type of platelet organelle, the dense body58 ( Fig...
متن کاملA platelet abnormality in the Chediak-Higashi syndrome of man.
Platelets from two probands homozygous for the Chediak-Higashi syndrome have approximately 10% of the normal number of serotonin-containing dense bodies as visualized electron microscopically in air-dried whole mounts. Since transport of serotonin across the platelet plasma membrane proceeds at a normal rate, and the few dense bodies present appear to store normal amounts of serotonin, the abse...
متن کاملThe Macromolecular Paracrystalline Lattice of Insect Viral Polyhedral Bodies Demonstrated in Ultrathin Sections Examined in the Electron Microscope
Thin sections of polyhedra obtained from gipsy moth larvae infected with P. dispar virus and from silkworm larvae infected with B. mori virus revealed viral particles contained within a pseudohexagonal, macromolecular, paracrystalline lattice. The gipsy moth virus occurs in bundles of one to eight rods enclosed by a limiting membrane. The particles of the silkworm virus, although generally occu...
